    摘要:

    昆虫信息素诱导植物抗性的现象已在多个研究体系中得到证实,对筛选植物抗性诱导剂以抵御有害生物具有重要意义。通过探究云杉八齿小蠹聚集信息素处理红皮云杉后对病原菌波兰长喙壳菌定殖的影响,旨在筛选出能够诱导红皮云杉抗波兰长喙壳菌的化学物质。采用注射法将云杉八齿小蠹聚集信息素的主要成分2-甲基-3-丁烯-2醇和S-顺式-马鞭草烯醇导入红皮云杉韧皮部。在处理后的第1天和第30天分别接种致病菌波兰长喙壳,并在接种后第60天统计波兰长喙壳菌在红皮云杉韧皮部的病斑大小。同时分析病健交界部位单萜烯的含量,评估其对波兰长喙壳菌生长的影响。结果表明,云杉八齿小蠹的两种聚集信息素成份处理后1天的红皮云杉不能抑制波兰长喙壳菌的病斑大小,但2-甲基-3-丁烯-2醇处理30天后显著抑制了波兰长喙壳菌在红皮云杉上的定殖,其诱导红皮云杉对波兰长喙壳菌的抗性与植物激素茉莉酸甲酯激发的抗性效果差异不显著。在信息素处理组中,仅2-甲基-3-丁烯-2醇处理红皮云杉30天后,与波兰长喙壳菌互作后可以激发寄主产生更多的单萜烯,室内生物测定结果显示单萜烯可以抑制波兰长喙壳菌的生长。研究证明了云杉八齿小蠹聚集信息素成分2-甲基-3-丁烯-2醇可以诱导红皮云杉对病原菌波兰长喙壳的抗性,经其长期诱导处理的红皮云杉与波兰长喙壳互作后防御性物质单萜烯明显上调是抑制该病原菌定殖和生长的关键因素。

    Abstract:

    The phenomenon of insect pheromones triggering host resistance had been well-documented across various research systems, which was important for identifying and screening plant resistance elicitors to counteract pest damage. This study examined the effects of treating Picea koraiensis with the aggregation pheromone of the Ips typographus on the colonization of the pathogenic fungus Endoconidiophora polonica. The primary goal was to identify chemical compounds that could induce resistance in P. koraiensis against this pathogen. The aggregation pheromones 2-methyl-3-butene-2-ol and S-cis-verbenol were injected into the phloem of spruce, and E. polonica was inoculated on the 1st day and 30th days after treatment. The lesion size of E. polonica in the phloem was measured at 60dpi, and the monoterpenes contents at the inoculation site were analyzed to determine their effect on fungal growth. Our results showed the aggregation pheromone treatment could not inhibit the lesion size of E. polonica in phloem of P. koraiensis after 1 day treatment. But 2-methyl-3-butene-2-ol treatment significantly inhibited the colonization of E. polonica in P. koraiensis after 30 days treatment, and the induced resistance of P. koraiensis to E. polonica was similar to that induced by the plant hormone methyl jasmonate. After treatment with 2-methyl-3-butene-2-ol for 30 days, the amount of total monoterpene increased significantly after interaction with E. polonica. Laboratory bioassay showed that all monoterpenes could inhibit the growth of E. polonica. The aggregation pheromones 2-methyl-3-butene-2-ol could induce the P. koraiensis resistance against pathogenic E. polonica. The up-regulation of the defensive substance monoterpene after the interaction between P. koraiensis and E. polonica was the key factor to inhibit the colonization and growth of this pathogenic fungi.
