现代北半球温带植物区系的地理分布和遗传格局等受新生代以来地质事件和气候变化的深刻影响[1-4]。北半球温带植物区系中物种多样性最高的是东亚中国-日本植物区系, 物种数约是面积和环境相似北美东部植物区系的两倍[5]。中国-日本植物区系的核心部分为中国亚热带地区, 即秦岭-淮河(34°N)与南部热带(22°N以南)之间的区域[6]。该地区作为世界生物多样性的热点地区, 既是中新世和上新世时演化谱系在更新世时重要的避难所, 也是植物丰富度和特有性程度极高的地区[7-9]。亚热带地区丰富的植物物种多样性与中新世以来复杂的地质气候历史密切相关。
中新世以来东亚主要经历的地质气候变化有全球变冷、青藏高原抬升以及亚洲季风和亚洲内陆干旱的形成和增强, 给植物的分布和演化带来了巨大的影响。第一, 全球气温在17—15 Ma(million years ago, 百万年前)经历气候适宜期, 小幅度变冷持续到上新世早期, 气温的持续下降迫使植物向南退缩。在渐新世晚期和中新世时高纬度地区连续分布的混交中生林即向南退缩, 在亚洲东部、北美东部、北美西部、亚洲西部和欧洲的东南部避难, 形成第三纪孑遗植物区系[2]。在上新世轻微回升后, 北半球冰期(3.6—2.4 Ma)将东亚的气候变成“冰窖”[10-11], 进入更新世。更新世时最典型特征为冰期和间冰期的周期性交替, 这种交替极大的影响了植物的分布[1, 3]。在冰期时, 植物向南退缩或向低海拔地区迁移, 在间冰期时, 植物向北扩张或向高海拔地区迁移。其中,末次冰盛期(23000—18000年前)对植物分布区的影响最大[1, 3, 12]。第二, 多项证据表明, 青藏高原自早中新世以来经历了4次抬升(22、10、3.6、1.1—0.6 Ma)[13-16]。作为海拔最高、地域最广的高原, 青藏高原是植物分区的自然边界和地理隔离的主要因素[17], 同时也是中国西高东低三级阶梯地形形成的重要原因, 自西向东复杂的地形结构使植物的分布在经度上存在明显差异[18]。尽管Renner[19]通过地质构造学、同位素、化石和气候模拟证据的收集, 对青藏高原抬升的时间及青藏高原在地质和气候带来的影响有了新的认识, 但无论哪种观点都认为青藏高原的隆起对植物分布和演化有着重要影响。第三, 亚洲季风在渐新世和中新世交界时(25—22 Ma)形成, 在15—13、8 Ma和3 Ma时增强[20-22], 在更新世时逐渐增强[23-24]。夏季季风给亚热带地区带来了足够的降雨, 改变了中新世前干旱的气候条件, 对遗传多样性增加有重要的影响[25]。亚热带西部地区(约105°E以西的高原地区)主要受印度季风影响, 东部主要受太平洋季风影响, 季风的不同可能与东西部不同的地形一起, 使西部地区的植物物种多样性比东部的低[26]。第四, 亚洲内陆干旱在渐新世和中新世交界时(26—22 Ma)开始, 在15—13、8—7、3.5、2.6、0.9—0.8、0.5 Ma时增强[21, 27-28]。与全球变冷一起, 亚洲内陆干旱也是植物分布区向南退缩的重要驱动力[29], 还可能作为气候屏障, 阻碍干旱带南北地区植物的基因交流, 触发东亚第三纪孑遗植物区系南北两个亚区的形成[30-31]。
通过地质气候变化对植物进化历史影响的研究, 可更好地理解该地区丰富植物物种多样性的起源和维持机制。近年来, 谱系地理学是主要的研究方法, 其重点是研究种内或近缘种内基因谱系的空间格局及其形成过程, 可探讨生物类群对地质气候变化的响应模式, 理解生物类群地理分布和种群分化的历史成因[32-33]。在2011年, Qiu等[26]对中国-日本植物区系中的谱系地理研究进行了综述。在亚热带地区, 通过落叶阔叶林植物银杏(Ginkgo biloba)、八角莲(Dysosma versipellis)和常绿阔叶林植物西藏红豆杉(Taxus wallichiana)、伞花木(Eurycorymbus cavaleriei)等的研究, 指出该地区阔叶林历史主要受更新世冰期间冰期的影响, 响应模式主要是局部地区高低海拔的迁移。自2011年以来, 有更多的研究阐述了地质气候变化对亚热带地区阔叶林植物进化历史的影响, 已呈现较清晰的谱系地理历史模式。以下两方面内容需要进一步总结, 以便全面了解该地区阔叶林植物的进化历史。一方面是更新时前地质气候变化对阔叶林植物分布区和谱系分化的影响, 很多植物的共祖时间可追溯到中新世, 则其进化历史不仅仅受更新世冰期间冰期交替的影响, 还受到中新世和上新世时地质气候变化的影响。另一方面是南退北进模式对遗传格局的重要性, 在更新世间冰期或冰期后, 并不是所有植物都在避难所周边经历局部扩张(localized expansion), 少部分植物存在明显的向北分布区扩张(range expansion)[34-35]。
总结近几年的谱系地理研究, 在中新世和上新世时, 由于全球变冷、青藏高原抬升以及亚洲内陆干旱的形成和增强, 阔叶林植物的分布区向南退缩, 在不同区域间形成不同的谱系, 形成分化。在更新世冰期间冰期的交替过程中, 阔叶林植物主要经历局部地区的收缩和扩张, 在多个避难所中存在长期隔离, 不同谱系进一步分化, 形成单系群, 形成较高的遗传多样性和遗传分化。少部分植物经历明显的向北分布区扩张, 南北走向的山脉为扩张路线。因此, 本文将首先介绍中新世和上新世地质气候变化对阔叶林植物分布区和谱系分化的影响。然后, 分别探讨落叶阔叶林和常绿阔叶林植物如何响应更新世冰期和间冰期的交替。最后, 从分化时间的准确估计和探讨遗传格局背后机制两个方面展望未来的研究趋势。
1 中新世和上新世时地质气候变化对阔叶林植物分布区和谱系分化的影响中新世和上新世是亚热带地区植物分布区变化和不同谱系形成的重要时期[2]。青藏高原抬升是中新世以来重要的地质事件, 不仅改变了地形, 还可能与亚洲内陆干旱和亚洲季风的形成与增强相关。亚洲内陆干旱与全球变冷一起, 迫使阔叶林分布区向南退缩到亚热带地区, 并触发不同谱系的形成。与此同时, 亚洲季风为亚热带地区带来了足够的降雨, 提供了有利的环境, 是遗传多样性增加的重要驱动力[25]。
首先, 中新世时地质气候变化迫使阔叶林植物向南退缩。在谱系地理研究中, 主要是使用叶绿体基因, 通过化石校准点估测分化时间, 从而建立进化历史与历史事件的相关性。如Kou等[25]使用叶绿体基因估算青钱柳(Cyclocarya paliurus)共祖时间在中新世中期(16.69 Ma), 表明气候适宜期(17—15 Ma)后的全球变冷可能迫使青钱柳从高纬度地区向南退缩到亚热带地区。Zhang等[36]使用叶绿体基因对光叶水青冈(Fagus lucida)和水青冈(F. longipetiolata)的谱系地理结构进行了比较, 认为两种植物的起源时间大约是6.36 Ma, 正好与全球变冷和亚洲内陆干旱增强的时间相一致, 全球温度的骤降和亚洲内陆干旱的增强可能迫使两种植物向南迁移[37]。其次, 中新世和上新世时地质结构变化触发了物种或种内不同谱系的形成。青藏高原在中新世间断性的抬升就是典型的地质事件, 抬升的山脉不仅使植物在地理分布上形成差异, 同时也可阻碍物种内的基因交流, 遗传结构和遗传多样性也会随之改变[38-40]。如水青树(Tetracentron sinense)、蓖子三尖杉(Cephalotaxus oliveri)、青冈(Cyclobalanopsis glauca)的谱系分化时间分别为9.6 Ma[41]、9.15 Ma[42]和9.07 Ma[43], 这些可能都与青藏高原在上述时间的地质结构改变相关。同样, 发生在上新世的青藏高原抬升与三叶崖爬藤(Tetrastigma hemsleyanum)的隔离事件[44]及珙桐(Davidia involucrata)西部谱系(四川盆地西部)的分化相关[45]。同时, 亚洲季风的形成和增强给中新世前干旱的亚热带地区带来了足够的降雨(现在年降雨量大于1000 mm), 为植物的生存和等位基因多样化提供了适宜的环境[25, 36]。水分是植物生长的必要条件, 影响了环境温度的调节和植物的扩散, 从而影响植物的进化历史。如水龙骨科瓦韦属(Lepisorus)植物在15 Ma左右经历了分化和首次扩张事件[46], 青钱柳则在9.6 Ma和3.6 Ma时有两次等位基因多样化增加的阶段[25], 均与亚洲季风增强的时间高度一致。
2 更新世时气候变化对阔叶林植物遗传结构的影响在更新世时, 亚热带地区虽没有大面积冰盖, 冰期间冰期的交替同样使气候产生了巨大变化。如末次冰盛期时, 气温比现在低4—6℃, 降雨量低400—600 mm[47-48], 这些变化对植物的遗传结构产生了深刻影响。通过化石孢粉数据可对植被分布模式进行重建, 在末次冰盛期时, 常绿阔叶林向南退缩至中国华南的沿海地区(24°N以南), 冰期后明显向北扩张至现在的纬度, 南部的常绿阔叶林(24°N以南)被热带森林替代。同时, 向北扩张的常绿阔叶林迫使落叶阔叶林向高海拔或者更高纬度的地区迁移[49-51]。
基于以上化石孢粉数据对植被分布的模拟, 常绿阔叶林和落叶阔叶林植物有不同的遗传预期。对于落叶阔叶林植物, 在冰期和间冰期时, 持续的长期隔离使其保持了较高的种群间遗传多样性和遗传分化, 种群内的遗传多样性会因遗传漂变而逐渐降低;高水平遗传分化使不同的谱系分布于不同地理区域中, 二次接触时在不同谱系间会形成接触带。常绿阔叶林植物则会保留向北扩张的遗传信号。南方避难所种群因更长的进化历史, 会有更多的古老和私有等位基因及更高的遗传多样性。冰期后扩张种群往往由避难所种群的子集构成, 因此有更低的遗传多样性和更多的衍生等位基因。除此之外, 在扩张的方向上, 连续的奠基者效应会使种群内遗传多样性逐渐降低[1, 3]。
2.1 落叶阔叶林植物的谱系地理历史自2011年以来, 越来越多的研究揭示了更新世时落叶阔叶林植物在多个避难所内原地(in situ)避难的模式[26]。在冰期间冰期的气候波动中, 隔离的冰期避难所种群在避难所周边经历周期性的收缩和扩张, 由于漂变的随机作用和瓶颈效应, 种群内部的遗传多样性逐渐降低, 种群间有限的基因流提高了种群间遗传分化和遗传多样性[52]。因此, 多个避难所假说预期低的种群内遗传多样性、高的种群间遗传多样性和高的遗传分化。在以往的谱系地理研究中, 遗传多样性模式与多个避难所假说的预期一致。首先, 在叶绿体基因中, 13个落叶阔叶林植物的种群内遗传多样性(within-population gene diversity, hS)的平均值(hS=0.180) 比总遗传多样性(total gene diversity, hT)的平均值(hT=0.782) 小的多(表 1), 总的遗传多样性(hT=0.782) 也比Petit等[60]汇编的170种植物的平均值(hT=0.670) 高。此模式可能也反映了该地区植物有较长演化时间[61], 因为在这13种植物中, 很多是第三纪孑遗植物。在核微卫星标记中, 种群内遗传多样性(observed heterozygosity, HO)同样比总的遗传多样性(total genetic diversity, HT)小, 如珙桐(Davidia involucrata)(HO=0.373, HT=0.747)[45]和瘦椒树(Tapiscia sinensis)(HO=0.513, HT=0.816)[59]。野核桃(Juglans cathayensis)为一个特例, 种群内遗传多样性(HO=0.682) 与总的遗传多样性(HT=0.782) 相似, 可能是有效的花粉基因流(风媒)阻碍了不同避难所种群间的分化[57]。其次, 遗传分化模式也符合多个避难所假说的预期。12个落叶阔叶林植物叶绿体基因遗传分化(inter-population differentiation, GST)的平均值(GST=0.799) 比Petit等[60]中的(GST=0.637) 高。微卫星标记中的遗传分化(inter-population differentiation, FST)同样很高, 如珙桐(FST=0.239)[45]和瘦椒树(FST=0.373)[59]。当然, 除了多个避难所相互隔离外, 植物间不同的生活史特征, 如雌雄异株的繁殖系统[62-63]及有限的花粉和种子扩张能力[64]等, 也会有助于高水平遗传分化的保持。

多个避难所假说认为局部扩张是间冰期时主要的种群动态历史[41, 54, 65-66]。然而, 近期研究发现部分植物经历了明显的分布区扩张。如木质藤本大血藤(Sargentodoxa cuneata), 分布在海拔130—2400 m的林缘灌丛中, 浆果由鸟传播[67]。Tian等[34]认为大血藤丰富的生境和有效的扩散方式使其在更新世时经历了多次分布区扩张。首先, 叶绿体单倍型的错配分析表明种群在95.98 ka(thousand years ago, 千年前)经历了一次扩张, 可能由末次间冰期时(75—100 ka)温暖湿润的气候触发。第三纪孑遗林冠树种青檀(Pteroceltis tatarinowii)也在该时期(90 ka)经历了向北扩张[56]。其次, 大血藤大多数叶绿体单倍型起源于0.63—1.2 Ma, 表明气候过渡期(mid-Pleistocene transition, 0.8—1.2 Ma)后冰期时的恶劣气候可能迫使分布区向南退缩, 为冰期后分化和向北扩张提供了大量的机会。祖先单倍型(H5) 周围星状(star-like)的单倍型网络图也支持气候过渡期后的扩张事件。最后, 多个避难所模式在亚热带地区已普遍存在。但是, 多个避难所间却共享着相同或密切相关的单倍型, 如大血藤中的H5, Gao等[68]中的单倍型H14和H18, Qiu等[66]中的单倍型H8, Lei等[54]中的单倍型H4, Zhang等[36]中的单倍型J等。在相互隔离的避难所中有祖先单倍型共享的现象看似相悖, 但存在两种可能的解释:长距离扩散和不完全的谱系分选。前者需要稀有的长距离扩散事件发生多次, 是一个可能性很低的情景。不完全的谱系分选则是更有可能的解释, 这种情景下, 祖先种群必然在破碎化前广布于亚热带地区, 祖先种群的扩张事件不可避免。多个避难所模式在大血藤中同样适用(私有单倍型H12、H18、H19分别分布在亚热带地区东部、西北和西南部), 估测的共祖时间(1.18 Ma)显示大血藤祖先种群的扩张事件可能发生在更新世中期。所以, 大血藤在更新世中期后经历了多次分布区扩张事件, 3条可能的扩张线路为武夷山-天目山、南岭-罗霄山脉-九岭山-大别山、南岭西部-云贵高原/南岭西部-三峡地区-秦岭[34]。因此, 亚热带落叶阔叶林植物的分布区扩张可能被低估了。
2.2 常绿阔叶林植物的谱系地理历史在谱系地理研究中, 与基于化石孢粉数据的植被重建不同, 亚热带地区的常绿阔叶林植物在更新世冰期时并没有完全退到南方避难所(24°N以南)中, 而是在预期南方避难所的北方(24°—33°N)存在多个“隐形避难所”, 即多个避难所模式[26, 69]。如葡萄科攀援藤本三叶崖爬藤, Wang等[44]通过遗传数据和潜在分布区模表明其在末次冰盛期时有多个避难所。首先, 种群内遗传多样性在研究区域内平均分布, 没有随纬度的增加而降低, 并不支持向北扩张假说[3, 70], 错配分析也没发现冰期后扩张信号。其次, 明显的谱系地理结构(叶绿体基因:NST=0.846 > GST=0.803, P < 0.05;核微卫星:RST=0.336 > permuted RST, P <0.01) 和地理隔离(isolation by distance)的缺失表明严重的“岛屿化”现象, 即种群间遗传分化主要由突变/漂变引起的, 而不是由基因流的减少/缺失造成[71], 不同“岛屿”的种群存在长期隔离, 在原地产生分化。第三, 叶绿体基因和核微卫星在西北、华中以及华南和华东谱系间相似的谱系地理隔离(phylogeographic break)也支持多个避难所模式。最后, 虽然生态位模型显示末次冰盛期时的分布区减少了约10%, 片段化也更严重, 但适宜的生境仍遍布整个分布区。因此, 三叶崖爬藤对冰期间冰期气候变化的响应符合多个避难所模式。虽然化石孢粉证据强调南方避难所的重要性[72-73], 但越来越多谱系地理研究发现中高纬度地区的北方存在冰期避难所, 揭示了北方避难所在冰期后群落动态中扮演着很重要的作用, 如鄂报春(Primula obconica)[74], 蓖子三尖杉[42], 栲(Castanopsis fargesii)[75], 青冈[43]等(表 2)。

在揭示常绿阔叶林植物符合多个避难所模式的同时, 在部分植物中也发现了南方大型避难所, 避难所种群在冰期后经历明显的向北扩张, 如优势种甜槠(Castanopsis eyrei)[35]和檵木(Loropetalum chinense)[76]。在甜槠中, 生态位模型显示末次冰盛期时主要分布在华南地区一条适宜的生境带中(约24°N), 与现在分布存在有限的重叠, 该生境带可能为南方大型避难所。核微卫星数据揭示了分布区西部和东部存在两个基因库(gene pool), 基因库中的遗传多样性都随着纬度的增加而显著降低, 表明现生种群是由两个南方避难所中的种群向北扩张形成。在两个基因库中还存在明显的亚结构, 表明存在多个“避难所中的避难所”[82], 不同区域中都有叶绿体私有单倍型分布也支持了多个避难所模式。在檵木中, 南岭地区同样是冰期南方避难所, 种群经两条路线向北扩张[76]。首先, 生态位模型预测檵木在末次冰盛期时适宜的生境位于华南和西南地区, 与古时候的植被分布模拟相一致[49-51]。南岭地区更高的遗传多样性、单倍型多样性和私有单倍型同样与南退北进模式的遗传预期一致[1, 3], 表明了南岭地区为末次冰盛期时檵木的南方避难所。南岭地区有很高的地形异质性, 保持了极高的物种多样性和特有性[9]。因此, 南岭地区受更新世冰期的影响很小, 是常绿阔叶林重要的避难所。在末次冰盛期后, 错配分析和BSP(bayesian skyline plot)分析进一步表明檵木种群在10.6 ka时经历了扩张。根据古老/衍生单倍型和遗传多样性分布, 南岭-武夷山和南岭-雪峰山-罗霄山脉为两条可能的扩张路线。当然, 与甜褚一致, 檵木在24°N以北也存在多个避难所(三峡地区, 大巴山和重庆省)。因此, 常绿阔叶林的谱系地理历史为:冰期时, 部分植物可退缩到南方大型避难所中生存, 同时, 北方复杂的地形和微环境可提供“隐形避难所”, 供残存的种群原地避难;在间冰期时, 避难所中的种群经历局部扩张或分布区扩张。
总结以上的研究, 常绿阔叶林植物的谱系地理历史与利用化石孢粉预测的植被分布模式并不一致, 而是与落叶阔叶林植物一致, 在亚热带地区的各大山脉中存在多个避难所, 如横断和大雪山脉、云贵高原、南岭和华中地区山脉等等。这些地区同时扮演古老谱系“博物馆”和新物种形成“摇篮”的角色[83], 在冰期和间冰期的交替过程中较小的气候波动有利于保持较高的植物物种多样性[84]。这些山脉同样可以作为谱系地理隔离, 阻碍不同谱系间的基因流, 使植物主要通过局部地区高低海拔的迁移来度过冰期间冰期的交替, 促进了不同遗传谱系间的进一步分化, 形成高的遗传多样和遗传分化。部分植物经历了明显的向北分布区扩张, 南北方向的山脉为扩散的廊道。在落叶阔叶林植物中, Tian等[34]认为大血藤经历了多次分布区扩张的原因是丰富的生境(海拔300—2600 m)和有效的种子扩散方式(以鸟传播)。而在常绿树种檵木和甜褚中, 两者都为分布海拔较低(海拔上限<1500米)的优势种, 种子也并没有有效的扩散方式(以重力传播)。因此, 尚不能总结出一般规律, 需要更多研究来总结经历明显向北扩张事件植物的生态学特征。
3 展望首先, 虽然已有部分研究中植物的共祖时间可追溯到中新世和上新世, 但仍相对较少, 且分化时间估测基本是基于叶绿体基因。叶绿体基因为母系遗传的单基因座遗传标记, 只可代表随机溯祖过程中的一次, 不完全的谱系分选会影响分化时间估测[85]。核基因则储存了更多的遗传信息, 如核微卫星可提供相互独立的多位点分析, 高度的多态性可精确推断种群的分化时间和分化历史[86]。随着测序技术的发展, 多位点的核单拷贝基因或全基因组信息也将更多的应用于谱系地理研究。同时, 相应的多基因座溯祖理论(multi-locus coalescent theory)必须要建立起来, 以解决多个基因座间不同的突变速率和迁移历史及基因重组等问题[87]。
其次, 亚热带地区阔叶林植物对历史地质气候变化的响应模式已较清晰, 但不同谱系间的显著分化和有限的基因流是因为地理隔离还是在不同环境中已形成适应性分化尚不清晰。生态分化种群间基因流的下降会提高中性标记的分化[88], 使得其与不同环境之间的差别存在显著的相关性, 即适应隔离(isolation by adaptation)[89]。要全面了解生态适应需要掌握两方面信息:形成适应性分化的特征和环境因子的相对重要性[90]。交互移植实验可判断适应性分化的强度, 推测适应性分化的特征和环境选择因子。通过实验植物在不同生活史阶段的性状表达和适合度之间的相关性可推测形成适应的性状, 通过环境因子和基因型相对性能之间的相关性可推测可能的选择因子[91-92]。在未来的谱系地理工作中, 探寻谱系地理格局背后的遗传机制也是一个发展方向。
