2009-2013年Bt棉田节肢动物群落多样性动态变化

摘要: 研究多年种植转Bt基因棉花对棉田节肢动物群落昆虫结构与组成、生物多样性的影响,明确其变化趋势,可为棉田害虫综合治理与生态调控、转Bt基因棉花环境安全性评价提供科学借鉴。于2009-2013年连续5年对转Bt基因棉花中棉所41和非转基因棉花中棉所49棉田节肢动物群落进行了系统调查,并结合5a气象因子的变化,分析了5年内棉田节肢动物群落相关参数的变化趋势。结果分析表明,2009-2013年中棉所41棉田昆虫群落、害虫亚群落和天敌亚群落所属目数有所上升,但差异不显著;所属科数、物种数、个体总数均呈现先上升后下降的趋势,个别年份差异显著,其余年份基本保持不显著的波动水平;昆虫群落和害虫亚群落多样性指数呈下降的趋势,至2013年下降达到显著水平,其余年份之间差异不显著,天敌亚群落多样性指数无显著变化;昆虫群落、害虫亚群落和天敌亚群落均匀性指数无显著变化;昆虫群落和害虫亚群落优势集中性指数有所上升,至2013年差异达到显著水平,天敌亚群落优势集中性各年份间无显著变化;与中棉所49棉田相比,相同年份中棉所41棉田昆虫群落、害虫亚群落和天敌亚群落结构与组成、多样性指数、均匀性指数和优势集中性指数均无显著差异。可见,短期内非剧烈天气变化对转基因棉田节肢动物群落、害虫亚群落和天敌亚群落在结构与组成、生物多样性方面没有明显的影响。

Dynamic changes in the biodiversity of arthropod communities in transgenic Bt cotton fields between 2009 and 2013

Abstract: In this study, effects of transgenic Bt cotton on the structure, composition, and biodiversity of arthropod communities were systematically investigated in cotton fields over five consecutive years, from 2009 to 2013. Non-transgenic cotton ‘CCRI 49’ was used as the control, and transgenic Bt cotton ‘CCRI 41’ was used as the test material. Effects of transgenic Bt cotton on arthropod communities provide an important theoretical basis for the comprehensive management of cotton insect pests and ecological control, as well as a scientific reference for the evaluation of the environmental safety of transgenic Bt cotton. In addition, sustainable development and use, which are in accordance with ecological succession, can be adopted on the basis of production and need for pest control in cotton plants. In this study, arthropod communities were systematically investigated in transgenic Bt cotton ‘CCRI 41’ and conventional cotton ‘CCRI 49’ fields, from 2009 to 2013. In addition, meteorological factors during these five consecutive years were considered when analyzing the changing trends of relevant factors in the cotton field arthropod communities. The results showed that, during 2009-2013, the number of "orders" in the insect community, pest sub-community, and natural enemy sub-community in the "CCRI 41" cotton fields increased in comparison with the control cotton fields, but the differences were not significant. The number of "families," "species," and "total number of individuals" showed an increasing trend over the first 3 years and then declined in the last 2 years (2012 and 2013). The differences were significant in some years, which might be due to the meteorological conditions, while fluctuations were within statistical limits in other years. The diversity indices of the insect and pest sub-communities showed declining trends, which were statistically significant in 2013 but not significant in the other years. There was no significant change in the diversity index of the natural enemy sub-community. In addition, there was no significant change in the uniformity indices of the insect, pest, and natural enemy sub-communities. The dominant concentration indices of the insect and pest sub-communities increased over the 5-year period, but the increase was only significant in 2013. There was no significant change in the dominant concentration index of the natural enemy sub-community in 2009-2013.There were no significant changes in the structure or composition of the cotton field insect, pest, or natural enemy sub-communities in Bt cotton ‘CCRI 41’ fields, nor were there any changes in the diversity, uniformity, or dominant concentration indices. It can be inferred from the results that there were no significant changes in the structure, composition, and biodiversity of the arthropod, pest, and natural enemy sub-communities in Bt cotton fields during the planting period. However, this study was a short-term comparative investigation of only one pair of cotton varieties. The long-term effects of planting transgenic Bt cotton on the structure, composition, and biological diversity of arthropod communities need to be investigated and monitored in the future. This will provide scientific data for a long-term safety risk assessment of transgenic Bt cotton.

生态系统生物多样性的变化是导致害虫重大变化的主要因素之一,棉田是受人为活动干扰、以棉花为中心的多种害虫、天敌共存、复杂网络的生态系统[1]。 棉花是我国重要的经济作物,棉花的生产受棉田害虫、天敌之间形成的节肢动物群落多样性的重要影响。棉田节肢动物群落多样性越高,其群落的稳定性越高,营养层之间能流越顺畅,棉田害虫受自然天敌的控制能力越强,棉花生产受到害虫的危害而引起的产量损失越少[2];另外棉田生态系统节肢动物群落多样性受气候、外界逆境(虫害、外源杀虫基因、干旱、涝渍)等多种因素的影响[3]。因此评价棉田节肢动物群落多样性的变化是国内外学者研究的焦点和热点[4-5]

外界因素及食物关系可引起昆虫群落内部害虫及其天敌种群动态变化,而昆虫群落的时空变化,会引起优势种发生演替,导致昆虫群落的多样性和稳定性也会发生变化[6-7]。棉田节肢动群落物结构与组成变化、生物群落不稳定,可引起某些害虫的大暴发,进而直接影响棉花生产,严重威胁着棉花的丰产丰收[8]。近年来,国内外学者对棉田节肢动物群落生态学进行了大量的研究[9-15],研究内容围绕转基因作物[16-18]、棉花生育期、棉田种植结构[19-20]及杀虫剂[21-23]等方面对棉田节肢动物群结构、种群动态、群落演替的影响等方面,这些研究极大的丰富了棉田节肢动物群落生态学和昆虫群落生态学的内容,但还没有得出一个明确的结果,表明不同影响因子对其生态系统的稳定性影响不同[17]。因此,转基因棉田节肢动物群落多样性长期跟踪监测可为转基因棉花环境安全性评价提供更可靠的理论依据。

因此,本文以非转基因棉花对照,本文从2009—2013年结合棉田气候变化,系统调查转Bt基因棉田昆虫群落、害虫亚群落和天敌亚群落的结构与组成、生物多样,明确其在中长期内种植对棉田节肢动物群落多样性的影响,以及如何影响等内容,为转Bt基因棉花环境安全性评价提出科学借鉴,并可根据生产和保护需要为棉田生态系统中昆虫群落的演替趋势、演替速度及演替方向等采取可持续发展和可利用的措施。

1 试验材料和方法 1.1 试验材料

中棉所49为非转基因棉花品种,简称常规棉;中棉所41为转Cry1Ac基因棉花品种,简称转Bt基因棉花;中棉所41和中棉所49不是同源亲本材料,但均是目前市场上出售且大面积种植的主栽棉花品种,均由中国农业科学院棉花研究所遗传育种研究室提供。

1.2 试验方法 1.2.1 试验设计

试验于中国农业科学院棉花研究所试验农场东厂进行(东经113°37′至114°58′,北纬 35°12′至36°22′,粘土类型)。每年4月28日—4月30日期间播种,棉花品种随机区组种植,小区面积不小于300m2,每品种3次重复。株距0.27m,行距0.8m。常规耕作管理,棉田全生育期不使用任何化学农药。

1.2.2 气象数据

2009—2013年由中国农业科学院棉花研究所试验农场安装的小型气象站监测所得。

1.2.3 害虫及其天敌昆虫调查方法

试验采用对角线五点取样方法(农业部953号公告-12.4—2007) [24],各处理棉田从5上旬开始至8月底,每5d调查1次2种棉田主要害虫及其天敌的种群数量。每小区每次调查10个样点,每点调查5株棉花。详细调查取样范围内地面和植株上昆虫的种类和数量,并作详细记载,记录所有直接观察到的节肢动物的名称、发育阶段和数量。开始调查时首先快速观察活泼易飞的昆虫和(或)蜘蛛数量。田间不易识别或者未知种类按统一编号进行记载归类。

1.2.4 数据分析方法

(1) 节肢动物群落的群落特征参数用物种丰富度、多样性指数、均匀度指数和优势集中性表示。物种丰富度(S)为群落中的昆虫种类数;相对丰度为占总丰富度的比例;多样性指数(H)用Shannon-Wiener多样性指数表示:,式中Pi为第i种物种个体数占群落个体数的比例;均匀度为实际观察的物种多样性指数,S为群落中的总物种数;优势集中性;应用Excel进行数据的初步计算和筛查,然后用SPSS 17.0软件对试验数据进行0.05水平多重方差分析。

(2) 变异系数

通常统计学方法是假设研究的空间变量为随机变量、相互独立的,通过计算研究变量的最大值、最小值、均值、标准差、方差和变异系数等来分析和研究变量的变异性。变异系数的大小反映了随机变量的离散程度,即表示所研究的变量变异性的强弱。其中变异系数的计算公式为:

式中,CV为变异系数(%),δ为标准差,μ为均值。

2 结果与分析 2.1 气象数据

2009—2013年1—8月河南安阳棉田相关气象资料如表 1所示。由表 1可见,2009年至2013年平均气温整体上呈上升的趋势,其中2011年最低,为24.8℃,2012年和2013年平均气温最高,均为25.6℃;2010年相对湿度最高,其次是2011年,2012年最低;降雨量2012年最高,达106.9mm,其次是2010年(96.5mm),2011年最低,为48.6mm。

2.2 两种棉田节肢动物群落结构与组成 2.2.1 两种棉田节肢动物所属目数

2009—2013年转Bt 棉田和非转基因棉田调查到节肢动物所属目数如表 2所示。由表 2可见,2009—2013年中棉所41和中棉所49棉田昆虫群落、害虫亚群落和天敌亚群落所属目数均有所上升,但变化差异均不显著,其中“中棉所41棉田”目数分别在10.7—12.7个、8.7—11.0个和6.0—8.0个之间,中棉所49棉田目数分别在10.7—13.0个、8.3—11.0个和6.0—8.0个之间;相同年份中棉所41与中棉所49棉田无显著性差异。表明2009—2013年试验期间,棉田昆虫群落、害虫亚群落和天敌亚群落所属目数变化不显著,转Bt基因对其无不利影响。

通过变异系数统计分析,两种棉田天敌亚群落的变异系数均较大,其次是害虫亚群落,表明年份间气候变化对天敌亚群落的影响较大。

2.2.2 两种棉田节肢动物所属科数

2009—2013年转Bt 棉田和非转基因棉田调查到节肢动物所属科数如表 3所示。由表 3可见,2009—2013年中棉所41棉田昆虫群落和天敌亚群落所属科数逐渐上升,其中2013年昆虫群落所属科数差异达到显著水平,害虫亚群落所属科数呈下降趋势,但差异不显著;中棉所49棉田昆虫群落在2011年上升到最大值,差异达显著水平,2012—2013年又有所下降,但与2009—2010年相比差异不显著;害虫亚群落呈上升趋势,2012年和2013年上升明显;天敌亚群落所属目数也呈增多的现象,其中2011年和2013年显著上升;相同年份转Bt基因棉田和非转基因棉田各营养层间无显著性差异。表明在2009—2013年试验调查期间,棉田昆虫群落、害虫亚群落和天敌亚群落所属科数有一定的变化。

变异系数统计分析表明,天敌亚群落在年份间变化差异较大,害虫亚群落变化差异小。

2.2.3 两种棉田节肢动物所属物种数

2009—2013年转Bt 棉田和非转基因棉田调查到节肢动物所属物种数如表 4所示。由表 4可见,2009—2012年中棉所41棉田昆虫群落和害虫亚群落物种数分别由63.7个上升至72.3个和35.0个上升至41.0个,差异均达显著水平,2013年均有所下降,但下降不明显;天敌亚群落物种数由28.7个上升到2011年的36.0个,差异达显著水平,2012年和2013年又有所下降,下降不明显。2009—2011年中棉所49棉田昆虫群落、害虫亚群落和天敌亚群落物种数分别由59.3个上升至77.3个、30.0个至40.3个和29.0个至35.0个,差异均达显著水平,2012年和2013年均有所下降,但下降均不明显。相同年份各营养层在转Bt基因棉田和非转基因棉田之间没有显著性差异。

变异系数分析表明,不同年份气候间棉田节肢动物群落物种数的变化差异来源主要是害虫亚群落,其次是天敌亚群落。可见不同年份间害虫亚群落间物种数变化波动性较大。

2.2.4 两种棉田节肢动物个体总数

表 5可见,2009—2013年中棉所41棉田昆虫群落和害虫亚群落个体总数无显著变化,天敌亚群落2013年明显减少;中棉所49棉田昆虫群落和害虫亚群落个体总数在2011年最高,差异达显著水平,其他年份差异菌不显著;各年份间天敌亚群落没有显著性差异;与中棉所49相比,中棉所41棉田昆虫群落、害虫亚群落和天敌亚群落个体总数在2011年分别下降了40.25%、41.6%和23.6%,前两者差异达显著水平,后者差异不显著。表明在多数年份转基因棉田和非转基因棉田昆虫群落个体总数没有显著性差异,但2013年转基因棉田昆虫群落的个体总数较非转基因棉田个体总数显著下降,主要是因为害虫亚群落个体总数显著下降,而害虫亚群落的显著减少可能由于平均气温、相对湿度和降雨量等单个因素或综合因素引起Bt棉外源蛋白表达量、营养物质、次生代谢物质等含量的变化,从而直接或间接引起食物链上害虫的数量变化。

变异系数分析表明,不同气候条件下,棉田昆虫个体总数变异差异主要来源于天敌亚群落,表明棉田昆虫个体总数的变化上天敌亚群落最敏感。

2.3 两种棉田节肢动物群落生物多样性 2.3.1 多样性指数

表 6可见,2009—2013年中棉所41和中棉所49棉田昆虫群落和害虫亚群落多样性指数呈下降的趋势,2013年下降达显著水平,其余年份间差异不显著;天敌亚群落多样性指数没有显著的变化;相同年份转基因棉田和非转基因棉田昆虫群落、害虫亚群落和天敌亚群落多样性指数无显著性差异。表明2009—2013年试验期间,两种棉田各营养层多样性指数无明显变化。

变异系数分析表明,不同年份棉田节肢动物群落多样性指数主要变异来源于害虫亚群落,与节肢动物群落物种数变异差异较大结论一致。

2.3.2 均匀性指数

2009—2013年转基因棉田和非转基因棉田节肢动物群落均匀性指数如表 7所示。由表 7可见,2009—2013年中棉所41和中棉所49棉田昆虫群落、害虫亚群落和天敌亚群落均匀性指数无显著的变化;相同年份转基因棉田和非转基因棉田昆虫群落、害虫亚群落和天敌亚群落均匀性指数无显著性差异。表明2009—2013年试验期间,两种棉田各营养层均匀性指数无显著变化。

变异系数统计分析表明,不同年份棉田节肢动物群落均匀性指数变异来源主要是害虫亚群落,天敌亚群落变异较小。

2.3.3 优势集中性指数

2009—2013年转基因棉田和非转基因棉田节肢动物群落优势集中性指数如表 8所示。由表 8可见,2009—2013年中棉所41和中棉所49棉田昆虫群落和害虫亚群落优势集中性指数有所上升,2013年上升显著;天敌亚群落优势集中性指数各年份间无差异;相同年份两种棉田昆虫群落、害虫亚群落和天敌亚群落优势集中性指数无明显差异。表明2009—2013年试验期间,两种棉田昆虫群落天敌亚群落优势集中性没有发生显著变化,害虫亚群落优势集中性指数增加,可能与近年来棉田刺吸性害虫种群数量显著增加,由原来的次要害虫上升为主要害虫有关。

变异系数统计分析表明,不同年份棉田节肢动物群落优势集中性指数变异主要来源于害虫亚群落,天敌亚群落变异较小。

2.4 棉田节肢动物群落表征参数与气候因子的相关关系

表 9可见,本文试验中5a平均气温、相对湿度和降雨量均与棉田节肢动物群落结构与组成、生物多样性无明显的相关性,表明棉田节肢动物群落的发生发展短期内与气象因子无显著的关系。

3 讨论与结论

生物群落结构比种群有更高的组织结构,内部具有复杂的多样性,它受时间、空间异质性、气候的稳定性、竞争、捕食与生产力等影响。表征生物群落的参数主要有多样性指数、均匀性指数和优势集中性指数等[25]。一般来说,群落结构越复杂,多样性指数就越高,群落的稳定性就越强。棉田易受棉花生产活动中,如棉花品种、农田布局、害虫防治、水肥管理等人为作业的干扰,再加上气象因素的影响,其生态系统也是一个多变的复杂生态系统。棉田生产活动在一定程度上影响棉田节肢动物群落的结构和动态,而节肢动物群落物种的组成与结构也会直接影响棉花植株的生长和生殖发育。

本文研究结果表明,2009—2013年转基因棉的中棉所41棉田和非转基因棉的中棉所49棉田昆虫群落、害虫亚群落和天敌亚群落所属目数有所上升,但变化差异不显著;所属科数、物种数、个体总数均呈现先上升后下降的趋势,个别年份差异显著,其余年份基本保持相当的波动水平,但试验期间与非转基因棉花相比转基因棉花对各项指标均无显著的影响,与Saxena 和 Stotzky等[26]、Baumgarte 和 Tebbe [27]、Marvier等[20]、 O′Callaghan等[19]、Hilbeck等[28]、Torres [29]低于3年的研究结果基本一致;两种棉田昆虫群落和害虫亚群落多样性指数呈下降的趋势,2013年下降显著,其余年份差异不显著,天敌亚群落多样性指数无显著变化;昆虫群落、害虫亚群落和天敌亚群落均匀性指数无显著变化;昆虫群落和害虫亚群落优势集中性指数有所上升,2013年差异显,天敌亚群落优势集中性指数各年份间无差异,和Torres等[29]、Thomazoni[30]的研究结果基本一致,虽然在年度间有部分差异,可能是由于年度间单个气候因素的变化或者多个气候因素的共同变化而引起;转基因棉田和非转基因棉田昆虫群落、害虫亚群落和天敌亚群落结构与组成、多样性指数、均匀性指数和优势集中性指数无显著性差异,该研究结果与Head [31]、O′Callaghan[19]、Whitehouse等[4]、Romeis等[32]的研究结果一致。

由于转基因生物风险特征决定了风险的复杂性,而目前转基因棉花对棉田昆虫群落多样性的安全性尚未达成一致的结论,因此外源基因的风险短期内较难判定。本文在根据前人1—3年期间的研究方法和研究内容,连续5a对转基因棉田和常规棉田节肢动物群落结构与组成、生物多样性进行系统调查,加长了调查时间,更能明确其发生发展趋势;同时对2009—2013年棉田节肢动物群落各参数与气象因子平均气温、相对湿度和降雨量之间的相关关系进行了分析,发现本文试验中短期内气象因子对棉田节肢动物群落无明显的影响,上述结论可为转基因棉花环境安全性评价提供理论依据,并为转基因生物安全管理部门提供必要的技术支撑。

本文在比较转基因棉田和常规棉田节肢动物群落结构与组成和生物多样性的试验过程中,棉田全生育期不施任何化学农药,在棉花害虫发生的一般年份不会对棉花生长产生毁坏性影响,但在棉花害虫大发生年份主要害虫(如棉铃虫、盲椿象、棉蚜等)会对棉花产量造成巨大影响,从食物链角度引起对棉田昆虫结构与组成及其生物多样性评价的偏差。因此在后期的评价试验中可增加棉田喷施化学农药处理为对照,同时在棉花大发生年份应根据具体情况结合天气因素进行化学防治,以保全棉苗及其正常生长,在后期数据分析和讨论中考虑其影响因素和程度。

文中对两种棉田昆虫群落、害虫亚群落和天敌亚群落进行了概括、比较和分析,明确了转基因棉花对其各指标的影响。但不同棉田不同昆虫(害虫和天敌)动态变化在转基因和非转基因棉田之间也可能会有差异,仅概括统计昆虫群落的动态变化可能会掩盖不同昆虫的差异,如抗虫棉田中的靶标害虫数量的下降,其他害虫如蚜虫种群数量可能会上升[33]。转基因植物环境安全评价不仅要明确其对群落演替性是否产生影响,还需明确该转基因植物对该生态系统中主要物种或稀有物种的影响程度,虽然该物种的变化不至于引起整个群落的演替,但根据其群落内部的细小变化可预测对群落稳定性影响的冲击力,从而预测该群落的稳定性。因此要了解转基因棉花对棉田主要物种或稀有物种是否有更细微的影响,需结合棉田小气候、农事操作等因素进行分析各物种在不同时期的变化。
