密度对大树蛙蝌蚪生长发育和存活率的影响

摘要:种群密度效应主要表现在影响和调节种群的死亡率、发育速率、繁殖率以及扩散、迁移等反应种群数量动态的重要参数.分析密度对大树蛙蝌蚪生长发育和存活率的影响,有助于探究外界环境因子导致大树蛙种群密度迅速下降的作用机制.设置了15、20、25 只/L和30 只/L 4个密度组,测量大树蛙蝌蚪的尾长、体长、发育时间和存活率.结果表明,在15-30 只/L范围内,密度升高显著降低了大树蛙蝌蚪尾长和体长的生长速率,减小其变态期的尾长和体长大小,其中与15 只/L相比,20、25 只/L和30 只/L 3组蝌蚪的尾长分别降低了11.6%、11.8%和13.9%,体长分别降低了11.1%、9.5%和12.9%;随着密度的升高,大树蛙蝌蚪发育至跗蹠部伸长期和前肢伸出期的所需时间显著延长;大树蛙蝌蚪生长后期的存活率随密度升高显著降低,但密度对蝌蚪生长早期的存活率影响不显著.因此,密度升高可显著减小大树蛙蝌蚪的尾长和体长、延长发育时间和降低其生长后期的存活率,可能影响大树蛙蝌蚪变态后的适合度.

The effect of rearing density on the development and survival of Rhacophorus dennysi tadpoles

Abstract:The population density of amphibians can greatly influence the dynamics of these populations by changing several key population parameters, such as mortality, development, reproductive rate, and dispersal and migration, especially in the larval stage. Meanwhile population density may change dramatically due to changes in the environment,, thus affecting the development and survival rate of the larval stage. Therefore, studying the impact of population density on larval growth and survival may help to understand the mechanism of rapid amphibian population declining caused by environmental factors. Nonetheless, the effect of population density on amphibian larvae is very poorly understood. In this study, we conducted laboratory simulation experiments to investigate the effect of rearing density on the development and survival of Rhacophorus dennysi tadpoles. We measured the tail length, snout-vent length, development time, and survival rate of Rhacophorus dennysi tadpoles at four rearing density levels (15, 20, 25 and 30 ind/L) and significant differences in tadpole performance were observed between density levels -tadpoles reared at higher densities were significantly smaller at metamorphosis, had longer larval development time and lower larval survival at later stage compared to those reared at the lowest density. Compared to tadpoles reared at the 15 level, tadpoles reared at 20, 25 and 30 density levels was 11.6%, 11.8% and 13.9% less in tail length, respectively, and 11.1%, 9.5% and 12.9% less in snout-vent length, respectively. In addition, increased rearing densities had also significantly prolonged the development time of tadpoles and lowered the survival rate at later stages, although they did not affect the early survival of the tadpoles.

两栖类动物是动物资源的重要组成部分,其种群数量在全球范围内持续锐减,甚至一些种类已经灭绝[1]。国内外学者对于引起两栖类种群数量下降因素(如气候变化、生境破坏和环境污染等)做了许多的研究[2, 3, 4, 5, 6, 7, 8],但这些研究主要是定性分析,而很少从两栖动物生长特征的变化来探讨种群下降原因[9]。密度是影响蝌蚪生长发育的重要因素[10, 11, 12, 13, 14]。高密度可导致林蛙(Rana sylvatica)蝌蚪[13]、虎纹蛙(Rana rugulosus)蝌蚪[15]和绿纹树蛙(Litoria aurea)蝌蚪[16]生长速率下降,变态个体减小和延长发育时间。而Semlitsh和Caldwell则研究发现高密度导致掘足蟾(Scaphiopus holbrooki)蝌蚪缩短发育时间[17],施林强等研究结果显示高密度对泽陆蛙(Fejervarya limnocharis)蝌蚪存活率和变态体重影响不显著[18]。虽然有关密度对两栖动物蝌蚪生长发育特征的研究已有诸多报道,但结论不一,因此对于密度对两栖动物蝌蚪生长发育特征的影响尚需进一步深入。

大树蛙(Rhacophorus dennysi)主要分布于我国四川、贵州、安徽、湖南等地山区溪流边的森林内或稻田、水坑附近的灌木和草丛中[19],以昆虫为食,繁殖季节成蛙选择在水田、水塘或溪流平缓处上方的灌木或草丛上吐泡排卵,卵孵化成蝌蚪时,泡沫溶解蝌蚪掉入水中[20]。湘西是大树蛙分布的主要区域之一,但近年来由于湘西大规模修建乡村公路、旅游开发和建设农田水利设施等原因,致使森林面积迅速下降,大树蛙种群数量急剧锐减,湖南省已将其列为地方重点保护动物。湘西为典型的喀斯特地区,森林覆盖面积的减少将导致土壤涵养水能力下降。一旦降水减少,许多溪流断流,水潭干枯,致使部分可供大树蛙蝌蚪生长的水域面积缩减,引起蝌蚪在发育期内种群密度不同程度的升高。为了探究大树蛙蝌蚪期种群密度的升高对蝌蚪生长发育及存活率的影响,本研究在实验室内设置了4 个不同的培养密度,研究密度因子对大树蛙蝌蚪尾长、体长、发育时间和存活率的影响,以揭示密度对大树蛙生长发育和存活率的影响,为进一步研究树蛙种群数量下降的机制提供参考依据。

1 材料与方法 1.1 实验动物

实验动物为孵化至第7 天的大树蛙蝌蚪。大树蛙成体(7雄,7雌)于2011年4月26日捕捉自湘西州吉首市跃进水库区溪流边的杉树,参照《中国两栖动物图鉴》鉴定[19]。分别配对(1雌1雄)饲养在2 m (L) × 1 m (D) × 1.5 m (H)的培养池,陆水面积各占50%,水深30 cm,陆地区栽种茶树和芋头供大树蛙休憩。每天18: 00给大树蛙投喂面包虫(Tenebrio molitor),每隔3d换1次水(pH值6.8),清理池中的排泄物。大树蛙抱对吐泡排卵后,任其自然孵化,孵化后的蝌蚪移至实验室饲养,室温控制在(24±1) ℃。

1.2 饲料

参照商业蝌蚪饲料营养水平[21],采用黄豆粉、玉米粉、麸皮、熟鸡蛋黄(自市场购买)、复合VB(湖北绿金子药业有限责任公司)和钙片(广西梧州制药股份有限公司)配置蝌蚪饲料,其100 g饲料的成分与营养水平如表 1

1.3 实验设计与饲养管理

雌雄配对的7对大树蛙3对排卵,为了消除亲代遗传、母体效应等影响,实验用的1080只蝌蚪均来自同一亲代,蝌蚪培养密度设置为:15、20、25 只/L和30 只/L,培养盆(直径×高 = 29.5 cm×9.0 cm)中水为2 L,水深约4.3 cm,每组设4个重复组和2个平行组(平行组用于补充实验组死亡的蝌蚪数)。每天12:00换水,清洗培养盆。每天换水之后投食1次,从15 只/L到30 只/L组的投食量分别为0.3、0.4、0.5 g和0.6 g,25 d之后,各组的食物量增加1倍(饲料组成及营养水平见表 1)。在室温(24±1) ℃,水温(20±0.2) ℃,光周期为12 L ∶ 12 D,培养蝌蚪至变态成幼蛙。

1.4 蝌蚪生长发育及存活率参数测定

每天记录蝌蚪的死亡数,移除死亡的个体,并从平行组中补充死亡的蝌蚪数。每隔6 d从每盆中随机捞取5 只蝌蚪置于水深1 cm的培养皿(直径×高 = 9.0 cm × 1.6 cm)中,进行拍照,采用ER Viewer 7.2软件处理照片,测定蝌蚪的尾长(TL)和体长(SVL)。当蝌蚪进入变态期,每天对蝌蚪进行多次观察并及时捞取出已进入变态期的蝌蚪(Stage 42)[22],待少量蝌蚪陆续进入变态时,停止测量蝌蚪形态参数。分别统计70%的蝌蚪发育至跗蹠部伸长期和前肢伸出期所需的时间[23]。继续饲养至所有实验个体均变成幼蛙时实验结束。

1.5 统计分析

采用协方差分析检验(General Linear Model)不同密度组蝌蚪的尾长、体长的显著性;体长与尾长的增长率采用反正弦转换数值后,用单因素方差分析(One-way ANOVA)显著性。采用单因素方差分析(One-way ANOVA)检验不同密度组蝌蚪发育时间的显著性。采用卡平方(X2)检验不同密度组在不同测定点存活率的显著性。所有统计分析在SPSS 13.0上进行,描述性统计值以平均值±标准误差(Mean ± SE)表示。统计显著水平为0.05。

2 结果 2.1 密度对大树蛙蝌蚪生长的影响

密度升高可显著减小蝌蚪变态期的尾长(F = 18.225,P = 0.0005)和体长(F = 11.010,P = 0.0004) (表 2,表 3),其中与15 只/L相比,20、25 只/L和30 只/L 3组蝌蚪的尾长分别减小了11.6%、11.8%和13.9%,体长分别减小了11.1%、9.5%和12.9%,但20、25 只/L和30 只/L 3组间蝌蚪变态期的尾长和体长无显著性差异(TL: F = 3.492,P = 0.370; SVL: F = 0.822,P = 0.445)。

蝌蚪尾长和体长的生长速率随密度的升高显著降低(TL: F = 38.112,P = 0.0003; SVL: F = 21.432,P = 0.0003),但25 只/L与30 只/L组间的体长和尾长的生长速率无显著性差异(TL: F = 3.171,P = 0.083; SVL: F = 1.220,P = 0.276)(图 1)。

2.2 密度对大树蛙蝌蚪存活率的影响

蝌蚪培养的前37 d内,密度对蝌蚪存活率的影响不显著(X2= 4.034,P = 0.258),但从第37 天至第67 天实验结束,密度的升高则显著降低大树蛙蝌蚪的存活率(X2= 2.479,P = 0.047)(图 2)。至67 d实验结束,15、20、25 只/L和30 只/L的蝌蚪或幼蛙存活率分别为82.5%、81.3%、75.5%和70.4%,表明蝌蚪存活率随密度的升高呈降低趋势。

2.3 密度对大树蛙蝌蚪发育时间的影响

随着密度的升高显著延长蝌蚪发育至跗蹠部伸长期(F = 23.090,P = 0.0002)和前肢伸出期(F = 30.023,P = 0.0007)的所需时间(表 4)。其中,30 只/L与15 只/L的蝌蚪发育时间相比,前者比后者蝌蚪发育至跗蹠部伸长期和前肢伸出期的时间分别延长6.7 d和6.2 d。

3 讨论

生存空间一定时,种群密度的升高会减小蝌蚪的生存空间,导致种内竞争加剧,从而影响蝌蚪的生长发育及存活率[24, 25]。本研究结果表明,密度升高会显著降低大树蛙蝌蚪的生长速率,减小蝌蚪变态期的尾长和体长大小,但20、25 只/L和30 只/L 3组间的蝌蚪达到变态期的个体大小无显著差异,却会显著延长蝌蚪变态所需时间。此与大多数的研究结论不同,即随着密度的升高,绿纹树蛙(Litoria aurea)蝌蚪[16]、泡蟾(Physalaemus pustulosus)蝌蚪[26]的变态个体逐渐减小。

种内竞争加剧引起蝌蚪之间的物理性接触增多,生存压力升高,为了获取足够的食物和氧气,蝌蚪的活动性增强,从而导致其生长速率降低[27]。Petranka发现豹蛙(Rana pipiens)蝌蚪在种内竞争加剧的情况下,生长快速、个体较大的蝌蚪会分泌抑制激素降低小个体蝌蚪的生长速率[28]。本研究发现25 只/L与30 只/L组内蝌蚪相对其他密度组的蝌蚪活动性更强,且随着密度的升高,大树蛙蝌蚪的生长速率显著降低,研究结果与上述推论基本一致。此外,本研究发现大树蛙蝌蚪生长后期的存活率随着密度的增加显著降低,可能是由于种内竞争加剧,引起生存空间的缩小和溶氧量的过度消耗从而导致了死亡率的增加。蝌蚪死亡率的增加,在一定程度上既可以缓解种内竞争,同时死亡的蝌蚪还可成为其他蝌蚪的食物[29]。Pfennig发现掘足蟾蝌蚪在生存环境变得恶劣的情况下,会从杂食性转变成肉食性的蝌蚪,肉食性的蝌蚪与杂食性的蝌蚪相比,前者拥有更高的生长速率,能缩短进入陆生阶段的变态发育时间,从而提高幼体的存活率[29],可能是掘足蟾蝌蚪对环境改变适应的一种生存对策。

蝌蚪的变态大小与变态时间影响其变态后的适合度[30, 31],而蝌蚪变态期个体大小除了跟本身遗传基础相关之外,主要与其生长速率和发育时间有关[17, 32, 33]。Newman[33]研究发现,在适宜的环境中,掘足蟾蝌蚪的生长速率高,发育时间短,变态个体大。而Semlitsh研究表明,高密度会抑制掘足蟾蝌蚪的生长速率,诱导蝌蚪变态发育,缩短蝌蚪的发育时间,但会减小蝌蚪的变态个体[17]。本研究表明大树蛙蝌蚪在20、25 只/L和30 只/L 3组密度处理条件下,大树蛙蝌蚪的生长速率显著降低,发育时间随密度显著延长,但实验个体达到变态期时尾长和体长无显著差异,此研究结果与施林强等[18]研究泽陆蛙的结果(高密度显著降低泽陆蛙蝌蚪的生长速率,延迟发育时间,但不影响其变态个体大小)基本一致,分析其原因,可能是高密度对蝌蚪变态延迟的一种补偿。但蝌蚪变态延迟有可能会持续影响大树蛙蝌蚪变态后的适合度,因此,有必要进一步探究密度对大树蛙变态后的生长发育及存活率。同时在以后的研究工作中,应设置更低密度并结合食物、温度等因素对大树蛙生长发育进行研究,有助于进一步阐述外界环境因素对大树蛙蝌蚪生长发育的影响机制。

综上所述,密度升高显著降低大树蛙蝌蚪的尾长和体长,延长大树蛙蝌蚪发育时间,降低其存活率。在湘西城乡建设、旅游开发等方面,应尽可能减少森林砍伐,减缓野生动物栖息地破碎化进程,对于保护两栖类濒危物种具有重要意义。
