在协同进化过程中,植物-食果动物之间形成了 较稳定的互惠关系[1, 2, 3] 。食果动物包括种子传播者 和种子捕食者,种子捕食者如昆虫、真菌和部分脊椎 动物,以种子为食物而缺乏传播作用,植物对此类捕 食者有防御作用[3] 。种子传播者多为脊椎动物,主 要包括鸟类、灵长类、翼手类等,通过取食果肉获得 食物;作为回报,传播者多以体内传播的方式将种子 搬运至新的地点,实现植物种群的扩散[4, 5] 。
植物次生物质是植物次生代谢过程产生的一系 列小分子化学物质,亦称植物次生代谢物(PSMs), 是植物与其它类型生物在长期演化过程中产生的。 国内外学者对植物茎、叶内次生物质的一系列研 究[6, 7, 8, 9] 表明,食草动物对植物茎、叶的取食压力,通常 促使植物茎、叶等部位产生较多次生物质防御捕食; 而次生物质又反过来潜在影响动物的取食行为、种 间关系和分布[10] 。因此,对植物次生物质的研究是 目前动、植物关系与生态系统中生物多样性维持机 制研究的热点之一。
从依赖脊椎动物传播的肉质果角度看,肉质果 营养丰富,季节性限制较强,具有强吸引性和短时效 性[11] ,除了在形态结构、物候等方面抵御食果动物 的捕食外[3] ,植物如何在挂果期内既能吸引动物传 播者取食果实传播种子,又能防御种子捕食者过度 消耗种子? 果实内的次生物质是否在此过程中起到 至关重要的调控作用? 尽管国内在非肉质果内次生 物质对植物-啮齿类互惠关系的调控方面做了大量 工作[12, 13, 14] ,但涉及肉质果内次生物质对植物-食果动 物之间的研究还远远不够[15] ,国外在这方面研究却 很丰富。鉴于此,本文从脊椎动物传播植物肉质果 中的次生物质类型、含量变化规律、生态作用等方面 展开论述,并提出未来研究需要关注的方向,以期能 促进植物与动物之间种子传播和互惠关系的深入 研究。
1 果实中次生物质类型植物次生物质种类繁多,根据次生物质的基本 结构特点,Dixon[16] 将其分为萜类化合物、酚类化合 物和含氮化合物三大类。很多依赖脊椎动物传播的 肉质果实中含有与植物茎、叶内相同的次生物 质[17] 。这些次生物质通常在果肉内分布居多,果皮 和种子(仁)中也可能同时含有。近些年,对依赖脊 椎动物体内传播(主要是鸟类)的茄属、辣椒属、樱桃 属等肉质果的研究表明,果肉内的次生物质类型丰 富,包括3 大类7 个类型(表1)。

肉质果中次生物质的含量随果实成熟而发生变 化。许多学者认为:未成熟果实中通常富含次生物 质,可以保护未成熟种子不被潜在的捕食者和食果 动物取食,而大多数次生物质在果实成熟过程中逐 渐递减,从而在种子成熟后促进食果者对果实进行 取食[15, 35, 37, 38] 。葡萄(Vitis vinifera)中酚类化合物浓 度在果实发育过程中的变化就表现出明显的规律。 果实从形成初始,直到果实转熟颜色转变时,单宁的 浓度随着果实的增重而增加,但颜色刚转变的果实 对于鸟类适口性较差;在果实进一步成熟、体积增大 的过程中,单宁浓度逐渐下降[30, 31] ;而葡萄中花青素 在果实颜色转变时就开始积累,在成熟过程中浓度 不断增加,直到果实发育后期浓度才有所下降[33] 。 Tsahar 等[28] 定量分析了地中海沙棘(Rhamnus alaternus)果实在成熟前后大黄素浓度变化与种子捕 食者对种子破坏的相关性。大黄素浓度在果实成熟 过程第1 阶段内上升,在成熟前达到1 个高峰,直至 果实成熟时大黄素浓度才降低到最低限度。在沙棘 果实逐渐成熟第一个月份里,大黄素浓度与无脊椎 动物或微生物捕食种子之间存在显著负相关,与其 它含有低浓度大黄素的植物果肉相比,高浓度种子 受损更少。这验证了多数学者的看法,即未成熟果 实中的次生物质能减少种子成熟之前被捕食的可能 性,在果实和种子成熟后这些次生物质会弱化或 消失。
但需指出,很多肉质果实植物在种子成熟后很 长时间内其果肉和种子中也会存在次生物质,甚至 一些次生物质的含量随着果实成熟而逐渐增多。 Estrada 等[39] 研究发现,辣椒属植物五彩椒 (Capsicum annuum) 的果实在发育早期(开花后14 d)即能检测到辣椒素,且其在开花21 d 后依然保持 较低含量,但在开花28 d 后辣椒素含量逐渐增加,在 辣椒发育最后阶段(开花后42 d)辣椒素水平达到最 高;游离酚的积累与辣椒素完全相反,发育早期的含 量远远高于发育的最后阶段。肉质果内的一些挥发 性次生物质,如番茄(Solanum lycopersicum)内的脱辅 基类胡萝卜素(Apocarotenoids)在果实成熟末期才释 放,在果实成熟过程中,脱辅基类胡萝卜素的含量可 增至40 倍[40] ;成熟草莓(Fragaria ×ananassa)中大多 数含硫挥发物的含量是未成熟草莓中的2 倍多[41] 。
3 果实中次生物质的生态作用 3.1 防御作用有学者曾提出关于成熟果实内次生物质对脊椎 动物防御作用的两个对立假说: 淤专毒性假说 (Directed Toxicity Hypothesis),即成熟果实中的次生 物质具有选择性,对非种子传播的食果实者毒性大, 但对种子传播者没有或者有极少毒性。于泛毒性假 说(General Toxicity Hypothesis),即果实中的次生物 质没有选择性,对所有食果动物都有毒性,毒性模式 与消费者对植物潜在的好处无关[38] 。
很多学者对这两种假说的论证,主要从某类 (种)特定次生物质的防御作用是否针对种子捕食者 而展开研究的。很多研究支持专毒性假说,其中涉 及到的次生物质只针对种子捕食者,这类次生物质 主要是氰苷和辣椒素。Struempf 等[25] 发现樱桃属 (Prunus)和接骨木属(Sambucus)成熟果实中的苦杏 仁甙(Amygdalin,一种芳香族氰苷) 对雪松太平鸟 (Bombycilla cedrorum)无抑制作用,雪松太平鸟可以 在4 h 内消费5—6 倍足以杀死实验鼠剂量的苦杏仁 甙。相同的研究表明,辣椒中的辣椒素对哺乳类(如 啮齿类等种子捕食者)有强烈的毒性刺激,但是对传 播鸟类无抑制作用[42] 。Levey 等[22] 通过对2 种野生 辣椒(C.chacoense和C.annuum) 果实进行录像检测, 发现成熟果实仅被鸟类取食,野外啮齿类都躲避这2 种辣椒,不取食和贮藏种子;而且笼养啮齿类也都避 免取食成熟五彩椒种子[42] 和含有辣椒素的人造 果实[43] 。
然而,也有研究支持泛毒性假说,这类次生物质 不单针对种子传播者,对所有食果者皆有毒性,主要 是生物碱。Cipollini 和Levey[18] 给2 种种子传播者 和2 种捕食者投喂2 种不同配糖生物碱(GAs)浓度 的天然茄属果实,发现种子捕食者和传播者都喜欢 低GAs 浓度的少花龙葵(Solanum americanum)果实, 高GAs 浓度的北美刺龙葵(S. carolinense)果实对所 有动物均有抑制作用。他们在另一研究[44] 中,通过 记录雪松太平鸟对3 种含有不同澳洲茄边碱琢- solamargine(一种配糖生物碱)天然浓度人造果实的 取食量,发现太平鸟受到了各种浓度琢-solamargine 的强烈抑制,而且不同浓度产生的作用是一样的。
3.2 调节动物取食频次和数量果实中的次生物质能够有效调节食果动物的取 食行为。鸟类取食时往往被迫在较短时间内离开母 树,以避免一次过多摄入同种特殊的次生物质[38] 。 Barnea 等[35] 对山楂(Crataegus monogyna)、常春藤 (Hedera helix)、欧洲冬青(Ilex aquifolium)肉质果肉 中皂苷(Saponins)和类黄酮(Flavonoids)的研究中, 发现当不同鸟类取食同种果实时,它们在树上都花 费相似的较短的时间(1.3—5.3 min),而且每次只取 食很少数量的果实(4—6 个/ min)。这些发现都间 接证实了果肉内温和的有毒物质可能阻止鸟类在一 次取食中消费过多果实的假说。一些学者认为,母 树能通过这种策略选择性地获得较多的短期访问 者,可能会使鸟类传播相似数量的种子,在进化上这 会使果实植物更具优势[38, 45] 。鸟类虽较早离开母 树,减少了某种鸟类的果实摄取量,但是能提高种子 传播到远离母树某些地方的可能性。
3.3 抑制和促进种子萌发成熟果肉中的次生物质既能阻止传播前种子的 萌发,又能提高传播后种子的萌发[45] 。通过食果鸟 类在消化作用中对果肉的移除,促进萌发过程的开 始;或者通过传播后残留于种子中的次生物质,保护 种子免受其它种子捕食者的取食,使种子活性和种 子萌发得以提高。
种子萌发抑制剂很大程度存在于一些植物的成 熟果肉中。比如,成熟茄属果实常含有GAs,它们能 延迟或抑制其种子的萌发[46] ,甚至对通过鸟类肠道 的种子也有同样的效果[21] 。同样,Yagihashi[47] 等人 对花楸属植物(Sorbus commixta)的研究结果显示,种 子萌发抑制剂存在于果肉中,其种子必须需要鸟类 取食和消化后才能萌发。Tewksbury[21] 等在一些喂 养实验中,发现取食含有辣椒素食物的鸟类,排出种 子的萌发率往往比对照组要高,次生物质对种子活 性的积极影响也许来自其对微生物和无脊椎有害生 物的防御,而不是直接促进种子萌发。
3.4 改变种子在肠道的滞留时间果实中的次生物质能改变种子通过肠道的速 率,产生种子较快通过的轻泻剂效应[48] 或减缓种子 通过速率的便秘效应[19] 。
家鸡和旅鸫(Turdus migratorius)分别被强迫食 下含有浓度为3.7—37 mg/ kg 和0.07—70 mg/ kg 的 大黄素果实后,都产生了强烈的腹泻[49] 。相反,黄 臀鹎(Pycnonotus xanthorrhous)取食含大黄素(0.01% 食物湿重)的食物后,增加了排便期间的平均时间, 表现出便秘效应[29] 。辣椒素和GAs 也能影响肠道 里种子的通过。Tewksbury 等[23] 给小嘴拟霸鹟 (Elaenia parvirostris) 和弯嘴嘲鸫(Toxostoma curvirostre)2 种鸟类,喂有注射辣椒素和对照溶液的 非辛辣辣椒,记录鸟类直到最后一粒种子被排出的 排便时间,与对照组相比,辣椒素平均增加了两种鸟 类15%—20%的种子滞留时间。Wahaj 等[19] 发现给 雪松太平鸟投喂含有北美刺龙葵天然果实GAs 溶液 的人造果实后,与对照组相比,雪松太平鸟表现出明 显的便秘效果。
靠动物传播的植物可以通过控制传播者肠道滞 留时间,继而影响种子的活性和分布。种子以较快 速度通过脊椎动物肠道,可以减少肠道对种子活性 的负面影响[23] 。然而,便秘效应对鸟类来说是有益 的,因为鸟类会有更高的消化效率[50] 。从植物的角 度来说,传播者的便秘效应可能在相当长的一段时 间内使其肠道携带种子,这会增加传播者将种子带 到更远地方的可能性[51] ,也会影响种子沉积模式, 包括传播者排便次数、每次排便的种子数量、推测的 传播距离[19] ,提高种子果肉彻底分离的可能性,而 这对于那些果肉剥离后才能萌发的种子来说是至关 重要的[52] 。相对于那些移动较多的动物(如燕雀类 鸟类和蝙蝠)来说,果实被移动较少的动物(如小型 哺乳动物)取食传播时,植物更能从动物的便秘效应 中获利[45] 。
3.5 对动物传播者的吸引作用成熟肉质果实中的次生物质能提供食果者容易 识别的觅食线索(如颜色、气味和味道),这些线索与 其果实回报(如糖、脂质、蛋白质、维生素和矿物质) 有关[38, 46] 。食果者能否利用次生物质预测果实质 量,需要依赖于次生物质与果肉营养回报的相 关性[38] 。
视觉吸引剂包括如越橘属(Vaccinium)果实中的 花青素(水溶性红色、蓝色和黑色)和辣椒属果实中 的类胡萝卜素(脂溶性黄色、橙色、红色和棕色),这 些次生物质都能在视觉上引诱动物前来取食[34, 53] 。 Saxton 等[32] 对葡萄中单宁、花青素与灰胸绣眼鸟 (Zosterops lateralis)和乌鸫(Turdus merula)的取食关 系进行了探讨。当喂食含相同次生物质绿色和紫色 人造葡萄时(野外葡萄皆紫色且含较高次生物质), 夏季时2 种鸟类对这两种颜色的葡萄都没有偏好; 但秋冬季时灰胸绣眼鸟较多啄食绿色人造葡萄,可 能是因为绣眼鸟无法通过消化途径处理次生物质, 所以认为紫色是果实含有高单宁酸毒性的线索,尽 量避免取食紫色果实;而乌鸫只吞食紫色人造葡萄, 可能是因为其储备冬天代谢和营养的需要,而且乌 鸫整吞果实能较快排出或呕出种子,所以紫色葡萄 中次生物质对其影响不大。
除了视觉吸引剂,还有一些次生物质可以起到 嗅觉吸引剂的作用。果实味道和气味是多种化合物 的混合作用[54, 55] ,在某些特定条件下,甚至一种“不 好冶的味道(如涩味),如果与果实回报有关,都能成 为引诱剂[56] 。香柏酮在葡萄柚(柑橘属,Citrus)果 实的成熟过程中显著增加,这种化合物是成熟果实 含有臭味的主要原因,成为动物取食的引诱剂[57] 。 Borges 等[58] 的研究也指出,孟加拉榕(Ficus benghalensis)果实通过释放挥发性次生物质吸引食 果动物的取食,夜间果实主要释放脂类化合物引诱 蝙蝠对其种子进行传播,而白天果实主要产生萜类 化合物吸引鸟类取食。
3.6 综合作用脊椎动物传播肉质果内的次生物质并不单独表 现其中某一种作用,往往是综合多种效应于一身。 次生物质的进化是植物对多功能性选择的结果[59] 。 因为次生物质生产成本较高,选择压力可能导致这 些次生化合物能在不同环境下产生各异的功能或目 的,此外这种多功能性与它们作为化学防御的主要 作用不矛盾[60] 。同一种花青素可以充当花的引诱 剂、抗氧化剂、植物抗毒素和抗菌剂[61] 。茄属成熟 果实包含生物碱,有多种生物活性,包括萌发抑制作 用、对哺乳动物的轻泻剂效应、对鸟类的便秘效应、 对草食动物和更多其它动物的抑制性作用[46] 。辣 椒属成熟果实中的辣椒素也具有多种功能,既能防 御种子捕食者,又能对真菌和无脊椎昆虫起到抑制 作用[42] ,其对种子萌发和种子肠道滞留时间的影响 又可以进一步促进种子传播[24] 。很多生物碱还可 以同时表现出异株克生、抗菌和对动物的毒性效应。 因此,相同的次生物质可能采取多种行为模式的行 动去阻止不同的生物群体。
4 研究展望肉质果中次生物质对植物-食果动物之间的调 控作用是复杂的,然而对肉质果内的次生物质与脊 椎动物之间相互作用的研究还不够深入。目前研究 主要关注植物通过产生次生物质、次生物质在果实 成熟过程中的含量变化对捕食和传播者行为的调控 机理[18, 22, 23, 24, 28] ,而果实次生物质的产生与果实营养 成分相互关联,同时果实形态、挂果期长短等特征也 必然对这一调控过程产生影响[62, 63] 。果实内次生物 质通过改变果实颜色(如花青素)和释放味道(如挥 发性脂质化合物)在吸引种子传播者、防御种子捕食 者等方面已得到较多的探讨和关注[64, 65] ,这些次生 物质大多随着果实的成熟而增多,食果动物可以通 过视觉、嗅觉感知它们的变化,然而食果动物如何感 知随果实成熟而不断减少或消失的次生物质? 结合 果实营养、形态、挂果期长短等因素,食果动物又如 何对同果期植物果实进行选择? 这些都需要进一步 进行探讨。因此,未来研究需要综合考虑果实次生 物质与其它生理生化、形态学等特征对脊椎动物的 综合调控机理。
从肉质果实形成开始,果肉和种子内的次生物 质经历种子传播和种子萌发过程,次生物质在种子 传播后的调控作用对植物种群或群落结构和分布格 局的影响应受到重视。肉质果实植物虽无法回避食 果动物的取食,但其利用果实(种子)内次生物质,通 过影响食果动物的取食时间、取食频次、取食数量、 肠道内种子滞留时间(轻泻剂或便秘效应)等方式实 现调控。肉质果实通过食果动物肠道时,果肉被消 化,种子形态和生理发生了变化,可能影响传播后种 子的活性和萌发;种子被一次性大规模集中或多次 分散排出以及种子传播距离的远近,也可能改变种 子在传播后微生境下的存活率和萌发率,这势必对 植物更新起到一定的影响[66] ,甚至改变植物种群或 群落的结构和分布格局。被沉积的种子暴露在各种 生物有机体存在的环境中,种子内的次生物质如何 调控传播后的种子与种子捕食者和病原微生物的关 系,目前这方面的研究尚很匮乏[24] 。因此,从群落 或种群角度,研究肉质果实中次生物质对传播后种 子活性和萌发率、植物种群或群落结构和分布格局 的影响,以及次生物质对传播后的种子与其它生物 相互关系的调控作用都是今后研究的方向。
依赖动物传播的植物面临如何吸引种子传播 者,同时又排斥非传播捕食者的进化难题,次生物质 与此进化问题密切相关。从动植物协同进化的角度 探讨植物次生物质的产生、防御和吸引策略将是探 讨上述难题的一个思路,比如食果动物对果实的选 择压力如何促使果实内次生物质的产生;次生物质 的进化如何对脊椎动物的果实选择和消费产生影 响;同一进化地理条件下的远缘植物类群或不同进 化条件下具有亲缘关系的植物类群在次生物质含量 及其生态调控方面是否存在差异等。
作为21 世纪植物化学生态学值得关注的前沿 领域之一[67] ,植物次生物质与其它生物类群进化关 系的相关研究需要多学科各种领域的整合,比如植 物化学、植物学、动物学、微生物生理学、生态学、进 化生物学等。理解脊椎动物和果实之间相互作用的 性质,对互利共生物种间的保护有重要意义。未来 果实次生物质的研究将能开辟食果动物-果实相互 关系研究的新角度,有助于完善种子传播机制和动 植物协同进化理论,扩充动植物相互作用的理论 体系。
